The MK-7 form (Menaquinone-7) of vitamin K2 is derived from chickpea fermentation.

Vitamin K’s role in blood clotting is well known, but growing evidence shows it also plays other roles in the body, including maintaining bone health, preventing inappropriate calcification in soft tissues, and offering a protective role in inflammation. These processes are mediated by Gla proteins, widely distributed in tissues, which remain inactive unless carboxylated by vitamin K. These include a key bone protein, Osteocalcin, and another called Matrix Gla Protein (MGP). For blood clotting, vitamin K is essential in the liver synthesis of prothrombin (factor II) and clotting factors VII, IX, and X, which convert soluble fibrinogen into insoluble fibrin used directly in clot formation.

In terms of bone health, studies suggest that undercarboxylated osteocalcin, believed to regulate bone mineralization quality, is associated with low bone density and increased risk of fractures. Supplementing with vitamin K2 has been shown to reduce fracture incidence, slow bone mineral density loss, and improve bone strength.

In the circulatory system, recent studies show that Gla proteins can inhibit blood vessel calcification and even reverse it, restoring arterial flexibility—but this requires vitamin K2. MGP can prevent arterial and cartilage calcification in living animals. Lack of MGP in humans has been linked to serious pathological conditions. Recent research suggests that undercarboxylated MGP forms are associated with calcified areas in blood vessels.

The discovery that MGP is a key regulator of tissue calcification is especially important for diabetic patients, for whom vascular calcification is a recognized issue.

An intriguing observation is that two Gla proteins (C and S), essential for balancing bleeding and clotting, also have non-coagulation-related functions.

In its activated form, protein C has direct anti-inflammatory action.

A recent epidemiological study showed that higher vitamin K levels were associated with lower production of inflammatory markers. Protein S (and a related Gla protein called Gas6) are widely distributed in tissues.

They are believed to have broader functions in protecting cellular integrity, especially in the brain, making the structural integrity of these Gla proteins particularly relevant. Their discovery has sparked renewed interest in vitamin K research. It’s important to highlight that for all known Gla proteins, the Gla region is essential for biological activity. This means that in cases of vitamin K deficiency, their functions are compromised.
